I make a tube bead about the width of the particular size tile I want to make, minus a millimeter or so. The thickness of the tube is something you have to experiment with to get the right amount of glass.
Then I just put the tube in just the bottom of the press and push down a little to flatten it, and then turn it over and do the same thing on the other side. This gives you a lollypop shape.
To make sure the corners are filled in, I turn the lollypop sideways, and press a little to flatten each edge. This gives me a rough tile shape, and makes sure there is glass for the corners.
Reheat somewhat, and then press with both halves of the press.

I hope this makes sense to you. If not, I'll try to explain better.
